ELISA Kits
ELISA kits (enzyme-linked immuno-sorbent assay) are a common laboratory test to detect and quantify a target protein or small molecule. ELISA kits make use of interactions between antibodies, the target, and signal molecules for detection. The strength of the detection signal allows the user to quantify the amount of target in their samples.
Reddot Biotech offers ELISA kits in Sandwich or Competitive inhibition format.
In the sandwich ELISA format, the microtiter plate has been pre-coated with an antibody specific to the target antigen. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to the target antigen. Next, Avidin conjugated to Horseradish Peroxidase is added to each microplate well and incubated. When the TMB substrate solution is added to the wells, the colour change that occurs is proportional to the concentration of target antigen in the sample. The standard curve is used to calculate the concentration.
In the competitive inhibition ELISA format, the microtiter plate is precoated with monoclonal antibodies, or antigens, that are specific to the target antigen. This monoclonal antibody (or antigen) has ONE binding site. The antigen from your sample competes with an inhibitor antigen conjugated with biotin. If the biotin-conjugated inhibitor antigen complex binds to a monoclonal antibody, this interaction will produce signal. If the antigen from the sample binds to a monoclonal antibody, this interaction will NOT produce signal. Next, Avidin conjugated to Horseradish Peroxidase is added to each microplate well and incubated. The amount of bound horseradish peroxidase conjugate is reverse proportional to the concentration of the target antigen in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of the target antigen in the sample. The standard curve is used to calculate the concentration.
